چکیده

Introduction: Multi-echo T2 relaxation allows scientists to probe water micro-compartments in vivo [1]. T2 relaxation is sensitive to myelin content [2] and has uncovered previously undetected micro-compartments in MS and PKU pathological tissue [3]. We aim to characterize how T2 distributions of fixed brain change with different types of MS pathology, identified using ex vivo histological stains, in order to create a model of T2 distributions specific to stages of MS disease.

Untangling the R2* contrast in multiple sclerosis: A combined MRI-histology study at 7.0 Tesla

T2*-weighted multi-echo gradient-echo magnetic resonance imaging and its reciprocal R2* are used in brain imaging due to their sensitivity to iron content. In patients with multiple sclerosis who display pathological alterations in iron and myelin contents, the use of R2* may offer a unique way to untangle mechanisms of disease. Coronal slices from 8 brains of deceased multiple sclerosis patien...

Myelin water imaging in multiple sclerosis: quantitative correlations with histopathology.

Various magnetic resonance (MR) techniques are used to study the pathological evolution of demyelinating diseases, such as multiple sclerosis (MS). However, few studies have validated MR derived measurements with histopathology. Here, we determine the correlation of myelin water imaging, an MR measure of myelin content, with quantitative histopathologic measures of myelin density. The multi-com...

Imaging correlates of leukocyte accumulation and CXCR4/CXCL12 in multiple sclerosis.

OBJECTIVE To compare leukocyte accumulation and expression of the chemokine receptor/ligand pair CXCR4/CXCL12 in magnetic resonance imaging-defined regions of interest (ROIs) in brains from patients with chronic multiple sclerosis. We studied the following ROIs: normal-appearing white matter (NAWM); regions abnormal only on T2-weighted images (T2 only); and regions abnormal on T2- and T1-weight...

A gradient in cortical pathology in multiple sclerosis by in vivo quantitative 7 T imaging.

We used a surface-based analysis of T2* relaxation rates at 7 T magnetic resonance imaging, which allows sampling quantitative T2* throughout the cortical width, to map in vivo the spatial distribution of intracortical pathology in multiple sclerosis. Ultra-high resolution quantitative T2* maps were obtained in 10 subjects with clinically isolated syndrome/early multiple sclerosis (≤ 3 years di...

New rapid, accurate T2 quantification detects pathology in normal-appearing brain regions of relapsing-remitting MS patients

INTRODUCTION Quantitative T2 mapping may provide an objective biomarker for occult nervous tissue pathology in relapsing-remitting multiple sclerosis (RRMS). We applied a novel echo modulation curve (EMC) algorithm to identify T2 changes in normal-appearing brain regions of subjects with RRMS (N = 27) compared to age-matched controls (N = 38). METHODS The EMC algorithm uses Bloch simulations ...
